CJC-1295: One Dose, IGF-1 Elevated For Over A Week

In two randomized, placebo-controlled ascending-dose trials in healthy adults, a single subcutaneous dose of CJC-1295 raised growth hormone for six days or more and IGF-1 for nine to eleven days.

What was studied

Growth-hormone-releasing hormone works, but only for minutes. CJC-1295 is a GHRH analog engineered to bind serum albumin so that it circulates for days. Teichman and colleagues ran two small phase 1 trials to measure how long a single subcutaneous injection keeps GH and IGF-1 elevated, and whether repeated doses accumulate.

What the data shows

  • After one injection, mean plasma GH rose 2- to 10-fold for six days or more, in a dose-dependent way.
  • Mean IGF-1 rose 1.5- to 3-fold and stayed up for nine to eleven days.
  • The estimated half-life of CJC-1295 was 5.8 to 8.1 days.
  • With repeated dosing, IGF-1 remained above baseline for up to 28 days, with evidence of a cumulative effect.
  • No serious adverse reactions were reported; the authors singled out 30 and 60 µg/kg as the best-tolerated doses.

These are pharmacokinetic and hormonal endpoints in a small number of healthy volunteers. Body composition, strength and any clinical outcome were not measured. The compound is not an approved medicine.

Why it matters for research

This is the only peer-reviewed human dataset for CJC-1295 and the reason the molecule is described as a long-acting GHRH analog. It gives laboratories a validated time course for GH and IGF-1 response that any in-vitro or animal protocol can be benchmarked against.
