BPC-157: Faster Tendon-To-Bone Healing In Rat Models
In a rat Achilles detachment model, systemic BPC-157 improved functional, biomechanical and histological healing endpoints over 21 days, and countered the aggravating effect of a corticosteroid.

What was studied
Tendon-to-bone junctions heal poorly on their own. Krivic and colleagues at the University of Zagreb cut the rat Achilles tendon away from the heel bone and treated the animals once a day with the stable gastric pentadecapeptide BPC-157, with methylprednisolone, with both, or with saline. Dosing started 30 minutes after surgery and stopped 24 hours before each analysis point.
Healing was scored on several independent axes: a walking-based functional index, mechanical testing of the repaired unit, macroscopic appearance and immunohistochemistry of the repair tissue.
What the data shows
- Functional index values were substantially higher in BPC-157 animals at every timepoint tested.
- Load to failure, stiffness and Young's modulus of the healed tendon-to-bone unit were significantly increased.
- Repair tissue showed better organized collagen fibers, more collagen type I and a more advanced vascular pattern.
- Methylprednisolone consistently worsened healing; when combined, BPC-157 reduced that aggravation.
A follow-up paper from the same group (Inflammation Research, 2008; 57: 205–210) looked at the first four days in 72 rats and attributed the early functional gain to reduced myeloperoxidase activity and inflammatory cell influx together with an increased vascular index.
All of this work is in rodents and comes from one research group. No controlled human trial of BPC-157 has been published. The data supports a research hypothesis about tendon-to-bone repair, not a clinical claim.
Why it matters for research
The study is the reference model for BPC-157 in connective tissue work: it defines the surgical model, the dose range across six orders of magnitude and the endpoint set that later papers reuse.
